Where do I start in terms of creating sprites similar to the style of RO?

I read the stickied posts, but what are the most modern tools (I'm on OSX) and instructions for making sprites like that of RO with frame? I'm programming a mobile game in my freetime and love the artwork of RO.. Thus far I've been using the frames off of ratemyserver as placeholders, but I'd ultimately like to make something similar on my own. How difficult is this trek? Thanks
